Single Cell Imaging and Mass Cytometry Analysis Platform (SCIMAP)

Mass cytometry is a hybrid technique combining flow cytometry and mass spectrometry. It uses antibodies conjugated with heavy metals rather than fluorochromes to label cells expressing proteins of interest, followed by time-of-flight (TOF) mass spectrometry for detection. The advantages include significantly enhanced multiplexing without the need for compensation between channels.

The Single Cell and Imaging Mass Cytometry Platform (SCIMAP) is a core facility at the Rosalind and Morris Goodman Cancer Institute (GCI) housing Fluidigm CyTOF (Cytometry by Time of Flight) mass cytometry technologies for analyzing dissociated tissues and blood (Helios) and tissue sections (Imaging Mass Cytometry (IMC) - Hyperion), with dedicated, highly qualified personnel to assist with assay development and data analysis. In collaboration with Fluidigm and via in-house optimization, SCIMAP provides validated and customizable antibody panels for detecting immune cell lineage and activation status and other phenotypes, reducing optimization time and costs.

Instruments

Helios

Helios is the third-generation mass cytometer (CyTOF) produced by Fluidigm. It performs phenotypic analysis of single cells using inductively coupled plasma time-of-flight mass spectrometry (ICP-MS). This allows analysis of single cells in unprecedented depth and breadth with minimal sample sizes. More than 40 markers can be analyzed simultaneously on a single cell. This allows the capture of a tremendous amount of data from a relatively low number of cells.

Hyperion

The Hyperion (Fluidigm) performs imaging mass cytometry, a complementary technology to flow-based mass cytometry that allows analysis of tissue samples stained with metal-conjugated antibodies, obtaining spatial resolution of protein expression at the single-cell level. The tissue is scanned by a small-spot-size pulsed laser and 1-micron spots are vaporized on each laser shot and analyzed by mass cytometry. Metal isotopes associated with each spot are measured and indexed against their location in the tissue. This allows the mapping of target proteins throughout the tissue section and the digital reconstruction of the tissue section as well as the analysis of single cell parameters in the microenvironment. Tissues can be interrogated with more than 40 parameters simultaneously. The Hyperion provides the advantage of preserving information on antigen localization, generating rich datasets combining the expression and spatial localization of many antigens in an intact tissue sample.
