Single-molecule control and exquisite visualization of macromolecular dynamics and interactions enables my biophysics group to broaden our understanding of fascinating molecular processes occurring on nanoscale dimensions, relevant to the nucleus of living cells. One axis of my research pioneers customized nanoscale geometries to explore the influence of physical confinement on DNA and protein dynamics and topologies. A second axis uses molecular biology tools to explore how intrinsic aspects of DNA topology, such "supercoiling", regulate interactions with oligonucleotides and polymerases. A third axis leverages our custom instrumentation to unscroll the genome in new ways, in partnership with diagnostic industries.
