Biographie: 

Le Dr. Peter McPherson, Ph. D., MSRC, est professeur de neurologie et de neurochirurgie, ainsi que d’anatomie et de biologie cellulaire à l’Institut neurologique de Montréal (Neuro) de l’Université McGill. Il est titulaire d’une chaire James McGill de l’Université McGill et fait partie du Groupe de recherche sur les maladies neurodégénératives du Neuro. 

Le laboratoire de Peter S. McPherson se sert d'approches biochimiques, moléculaires, structurales, génétiques et cellulaires pour identifier et effectuer la caractérisation fonctionnelle des protéines qui interviennent dans la formation des vésicules à manteau de clathrine. Celles-ci sont les principaux véhicules de l’absorption endocytique de multiples charges protéiques et lipidiques, dont des nutriments et des récepteurs de signalisation. Après l’endocytose, la charge est livrée aux endosomes d’où elle est recyclée vers la surface cellulaire ou dirigée vers les lysosomes aux fins de dégradation. Ces décisions de tri contrôlent la localisation et les niveaux de protéines et sont altérées dans le cancer et la maladie neurologique. Ainsi, des projets en cours au laboratoire révèlent comment un dérèglement dans le transport de charges sélectives des endosomes à la surface cellulaire contribue au développement de glioblastome et du cancer du sein.

Auparavant, le laboratoire du Pr McPherson a eu recours à la protéomique subcellulaire (fractionnement subcellulaire et spectrométrie de masse à haut débit) pour identifier le complément intégral de protéines qui caractérisent les vésicules à manteau de clathrine de plusieurs tissus. Parmi les protéines identifiées figurent certaines qui n’étaient connues que sous forme de cadres ouverts de lecture non caractérisés. Le laboratoire du PrMcPherson a caractérisé la fonction de certaines des nouvelles protéines, mais il en reste un nombre important à étudier. Une protéine récemment identifiée contient un module appelé un domaine DENN. Le Pr McPherson et ses collègues ont démontré que les domaines DENN fonctionnent de façon enzymatique comme un facteur d’échange du nucléotide de la guanine pour activer de petites GTPases de la famille Rab. Le génome humain compte au minimum 26 protéines à domaine DENN et un champ important d’études au laboratoire porte sur la relation de ces protéines avec les ~70 Rab fonctionnant dans le trafic membranaire. Un exemple particulièrement frappant est la protéine à domaine DENN C9orf72, dont la fonction est inconnue. Une mutation dans C9orf72 est la cause la plus fréquente de formes génétiques de sclérose latérale amyotrophique (SLA) et le laboratoire cherche à comprendre la relation entre le trafic des membranes endosomiques et la pathogenèse de la maladie.

Certaines protéines identifiées ou étudiées au laboratoire du Pr McPherson ont été associées à des maladies neurologiques comme la maladie de Huntington, l’ataxie spastique récessive autosomique de Charlevoix-Saguenay (ARSACS) et la maladie de Parkinson (MP). Par exemple, nous avons démontré que le LRRK2, gène majeur de la MP, se lie aux clathrines et fonctionne dans le trafic des membranes endosomiques. De plus, nous avons montré que l’ARSACS a la même physiopathologie que la MP. En fait, des altérations dans la régulation du transport membranaire émergent comme un thème central des maladies neurodégénératives. Comprendre le fondement biologique cellulaire de la maladie neurologique est un nouveau champ d’intérêt du laboratoire.
